IOF will be demonstrating the Nikon Opti-MicroScan (UV-OMS)  in May/June  2024 at the Imaging & Optics Facility on ISTA campus.

Location: Berthalanffy Faundation building: room I04.U1.019

All tests need to be coordinated with IOF:  Please write to IOF@ist.ac.at if you are interested to obtain a test-slot.

The Nikon Opti-MicroScan (UV-OMS) is designed for photo-stimulation and photo-ablation experiments. The demo-system comes with a visible laser (405 nm laser on demo device) for photo-stimulation and a UV-pulses laser (355 nm) for photo-ablation, it enables a wide range of potential applications including photo-bleaching, photo-activation, photo-switch, photo-conversion, photo-ablation, photo-damage, irradiation, uncaging, and micro-patterning.

The UV-OMS will be mounted on a Nikon fluorescence widefield microscope Ti2-E, allowing for simultaneous ablation or photo-bleaching and imaging between 405-750 nm, potentially in many flexible configurations. Importantly, the Ti2-E fluorescence widefield microscope which is available for the demo includes standard filter sets and a light source for imaging common dyes like DAPI, CFP, FITC, YFP, TRITC, mcherry, and Cy5.

Key Features:

  • Simultaneous imaging and photo-manipulation
  • Fast rastering of user-defined patterns or sequences
  • Simple yet flexible setup with customizable power, wavelength, and dwell time for each pattern
  • Stimulation during live imaging on user request (mouse click)
  • Near diffraction-limited spot size
  • Full control using NIS-Elements software

The demo device comes with various objectives, including dry & water immersion (WI) objectives for dedicated live cell imaging experiments:

  • CFI Plan Fluor 40XC / 0.75 / 0.66 mm
  • CFI Plan Fluor 60XC / 0.85 / 0.35 mm
  • Apo 40x WI / 1.25 / 0.18 mm
  • Apo LWD 40x WI / 1.15 / 0.60 mm
  • PA VC 60x WI / 1.20 / 0.29 mm
